B1.1 Preactivated TentaGel® Resins

TentaGel® resins are grafted copolymers consisting of a low crosslinked polystyrene matrix on which polyethylene glycol (PEG or POE) is grafted. The PEG spacer is attached to the matrix via an ethyl ether group which increases stability towards acid treatment and minimizes PEG-leaching. As PEG is a „chameleon type“ polymer with hydrophobic and hydrophilic properties, the graft copolymer shows modified physico chemical properties which are highly dominated by the PEG moiety (and no longer by the polystyrene matrix). These graft copolymers are pressure stable and can be used in batch processes as well as under continuous flow conditions.
The PEG spacer is in the range of MW 3000 Da.

Aminomethylated polystyrene resins are prepared in-house from crosslinked polystyrenes by a direct aminomethylation reaction according to highly optimized protocols. Resins produced by our process are chemically defined, homogeneous and free from any additional crosslinking from the functionalization process. Aminomethylated polystyrene resins can easily be acylated with any appropriate carboxylic acid-containing linker using standard methods of amide bond formation to furnish supports for solid phase organic synthesis.

B1.2.2.1 HypoGel® -NH₂ Resins
"HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.
These resins show high loading and hydrophilicity between polystyrene and TentaGel®

B1.3.1.1.1 4-Formyl Phenoxy on HypoGel® Resins ( FP-linker)
HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.These resins show high loading and a hydrophilicity between polystyrene and TentaGel.
Due to the lack of amide bonds and of benzylic linkages as well as the very good swelling properties in a broad variety of solvents this resin is well suited for solid phase syntheses of secondary sulfonamides and carboxamides.

B1.4.2.1 HypoGel® amine acylated with succinic acid
HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.

B1.6.1.4 Merrifield Resin (Chloromethyl Polystyrene)
The Merrifield resin has in the past been the standard support for the synthesis of peptide acids by Boc strategy. Originally, the cesium salt of a protected amino acid was anchored to the chloromethyl support via nucleophilic displacement of chlorine. Although, Me4N salts, sodium salts in THF with Bu4NF catalysis and more recently zinc salts in EtOH have also been used. Cleavage is normally effected by treatment with HF or trifluoromethanesulfonic acid or by hydrogenolysis, alcohols can be released by using diisobutylaluminium hydride or LiBH4. For cleavage scavengers may be required. New isomeric trialkoxybenzylamine resins have been developed by coupling phthalimidomethyl-3,5-dimethoxyphenols to this resin followed by subsequent treatment with hydrazine, that allows DCC coupling with the carboxyl function of amino acids or peptides. The addition of alcohols can be performed by heating the corresponding potassium or sodium alkoxide and the resin in DMF. Reductive cleavage of the anchoring ester linkage was also accomplished.
Attachment of sodium dihydrocinnamate to this resin and treatment with a solution of lithium diisopropylamide (LDA) in THF formed a resin-bound zinc enolate for aldol condensations. Reductive cleavage with diisobutylaluminium hydride afforded a 1,3-diol library. The removal of b-lactams has been achieved by using aluminium chloride. Another application of this support to nonpeptides involved anchoring the cesium salts of bromo or iodobenzoic acids followed by subsequent transformation into biaryl derivatives using Suzuki coupling conditions. The products were released from the resin via nucleophilic attack with methoxide ion to give methyl esters.

B1.8.3.1 Polystyrene Ethyl Sulfonyl Chloride
This polymer bound alkylsulfonyl chloride resin is an equivalent to alkyl sulfonic chloride and can be used in numerous applications. When this resin is treated with nucleophiles, a variety of polymer bound sulfonates can be created ( e.g. sulfonyl azides, sulfonyl hydrazine) and subsequently used as polymeric supports, polymer bound reagents or in catalytic organic synthesis. Application in the "catch and release" strategy, where the resin catches a compound to form a polymer bound tosylate. Purifying the polymer bound tosylate by washing and subsequent treatment by a nucleophile, releases the target product from the resin. A typical application is to form polymer bound tosylates by loading alcohols to the resin. Treatment with nucleophiles like amines, imidazoles or thiols generates the corresponding sec. or tert. amines, alkylimidazoles or thioethers.
